I am a medical genetist with a traditional interest for research activities related to diabetes and metabolic diseases, both from the clinical and the molecular point of view. During my first stage at the Diabetes Branch of the National Institutes of Health in Bethesda (USA), as a visiting associate under the guidance of Dr.. Jesse Roth and Dr. Simeon I. Taylor, I had the opportunity to demonstrate similarities between the insulin receptor and proteins encoded by the src oncogene (Science 227: 761-763, l985) Since then I cultivated an interest for insulin signaling and the relation between hyperinsulinemia sodium retention, cell proliferation and cancer. During a second stage at the same Diabetes Branch of the NIDDK, NIH, as a visiting Scientist under the guidance of Dr. Simeon I. Taylor I focused my attention of Sgk1 kinase activity as possible mechanism linking insulin action and sodium retention. This interest has been further pursued in collaboration with prof. Bonnie Blazer Yost. More recently, in Italy, I further defined the role of sgk1 in cell survival , proliferation and nucleo-cytoplasmic transport of MiRNAs. I also contributed , in collaboration with prof. Silvia schenone, to the identification of a specific inhibitor of Sgk1 that appears to be a potentially interesting drug in the treatment of several human cancers, when used alone or in combination with chemotherapy or radiotherapy. Preliminary data obtained again in collaboration with Prof. Bonnie Blazer yost and Prof. Silvia Schenone suggest that this inhibitor can be useful in the treatment of diseases associated with increased channel function like hypertension and hydrocephalus secondary to Meckel gruber syndrome. The possible use of Sgk1 inhibitors in the treatment of these channel diseases is the topic of the present grant submission.
